    Reaching the unreachable: new approaches to challenging drug targets

    Modern medicines act with remarkable precision. Antibodies, for example, recognise their molecular targets much like a key fits into its corresponding lock. However, certain targets like small molecules or carbohydrates remain difficult to target. The start-up Panosome aims to eliminate these blind spots with a novel immunotherapy platform that harnesses sophisticated masters of immune evasion: trypanosomes.

    A team led by researchers from the German Cancer Research Center, the HI-STEM* Stem Cell Institute, and Medical Clinic V at Heidelberg University Hospital has characterized T cells in the bone marrow in cases of multiple myeloma and acute myeloid leukemia: A specific group of tumor-reactive T cells can recognize and fight cancer cells, but is often not sufficiently activated.     Persistent infections with human papillomaviruses (HPV) are the primary cause of cervical cancer. Researchers from Heidelberg have developed a promising therapeutic vaccine consisting of immunogenic virus peptides linked to silica nanoparticles, which is currently being investigated in preclinical studies.     A cutting-edge chemical process is the first to make it possible to quickly and easily produce modified peptides with boronic acids. As part of this work, scientists managed to synthesize a large number of different biologically active peptide boronic acids and investigate their properties. They open up new possibilities in the young research field of synthetic immunology and could go on to be used primarily in immunotherapy.

The founders aim to introduce an effective strategy for combating solid tumours, with initial proof of concept targeted at prostate cancer.

    Antibody can improve immune cell therapy against leukemia

    Scientists from the German Cancer Research Center (DKFZ) and Heidelberg University Hospital (UKHD) have shown that the combination of therapeutic immune cells, known as CAR T cells, and a bispecific antibody could improve the treatment of leukaemia. In the culture dish and in mice, they tested CAR-T cells directed against the B-cell marker CD19 in combination with bispecific antibodies that bind to the B-cell-specific protein CD20.

    The Baden-Württemberg Ministry of Science, Research and Arts (MWK) is supporting medical researchers at Ulm University with start-up funding totalling 600,000 euros. The aim is to establish an EU consortium for the broad therapeutic use of mesenchymal stromal cells. The funding is being awarded as part of the "BEGIN - Participation in major European projects and initiatives" programme.

    Newly discovered mechanism of T-cell control can interfere with cancer immunotherapies

    Activated T cells that carry a certain marker protein on their surface are controlled by natural killer cells. In this way, the body presumably curbs destructive immune reactions. Researchers now discovered that NK cells can impair the effect of cancer therapies with immune checkpoint inhibitors in this way. They could also be responsible for the rapid decline of therapeutic CAR-T cells.
